Remove the cell processing bag and roll it around the rotating seal. Pass the cell
processing bag through the center hole of the centrifuge bowl cover (Figure
6-6 on page 6-7).

Prime the hydraulic system, as described in “Priming” on page 6-36.

Determine if processing will continue with the same cell processing set or if it
will be necessary to start over with a new cell processing set:
•
To continue processing using the same cell processing set, proceed to
step 17.
-or-
•
To start over with a new cell processing set, ensure that all cell product
has been returned to the cell product bag. Seal and remove the cell
product bag. Proceed to “Installing the Cell Processing Set” on page 6-2.

Roll the cell processing bag around the rotating seal and pass the cell
processing bag through the center hole of the centrifuge bowl cover.

Install the cell processing bag.

Replace the alignment blocks and the centrifuge bowl cover.

Close the sliding covers and the centrifuge cover interlock latch (if applicable).

Press the STOP RESET button.

Return to “Loading the Starting Cell Product” on page 6-14.
Excess Volume of Processing/Wash Solution
For protocols ending with resuspended cells, the Agitate/Wash-In operation must
be monitored to prevent the cell processing bag from being overfilled. If the cell
processing bag is overfilled, it is necessary to remove some of the processing/wash
solution in order to remove the centrifuge bowl cover.

Ensure the COBE 2991 is in Automatic mode.

Determine the volume of processing/wash solution that must be removed in
order to leave 450 mL or less in the cell processing bag.

Set the SUPER OUT VOLUME control to the volume of processing/wash
solution that must be removed.

Press the START SPIN button. The centrifuge spins, separating the cells from
the processing/wash solution.

When the cell product is adequately separated, press the SUPER OUT button.
The COBE 2991 advances to the Agitate/Wash-In operation when the volume
of supernatant expressed is equal to the setting on the SUPER OUT
VOLUME control.

When the COBE 2991 advances to the Agitate/Wash-In operation,
immediately press the STOP RESET button.
