Processing and Common Operations
Removing the Centrifuge Bowl Cover When the Cell Processing
Bag is Overfilled
If the COBE 2991stops with more than 450 mL of fluid in the cell processing bag,
it will be impossible to remove the centrifuge bowl cover because of friction
between the cell processing bag and the underside of the cover. This may occur if an
Excess Pressure alarm occurs during processing, or an excess volume of
processing/wash solution for resuspension is used. Depending on which condition
has occurred, use the appropriate procedure to remove the centrifuge bowl cover
and recover the cell product being processed.
• If an Excess Pressure alarm occurred during processing, refer to “Excess
Pressure During Processing”, below.
• If an excess volume of processing/wash solution for resuspension was used,
refer to “Excess Volume of Processing/Wash Solution” on page 6-33.
Excess Pressure During Processing
If an Excess Pressure alarm occurs during processing with no apparent cause, air
may be trapped in the hydraulic system, and the system must be primed. Before
priming, use the following procedure to remove the cell processing bag.
Note: If an
Excess Pressure
alarm occurs during
processing, DO NOT seal off
the cell product bag.
Remove an adequate
volume of fluid from the cell
processing bag allowing
removal of the centrifuge
bowl cover.

Ensure the sliding centrifuge covers are closed, the seal weight is in place, and
the centrifuge cover interlock latch is closed (if applicable).

Press the STOP RESET button.

Remove the clear tubing from the RCD.

Place a clamp on the purple-striped tubing (SOV).

Press the BLOOD IN button to open valve V1. Remove the red-striped tubing
from valve V1.

Place the cell product bag on the tabletop (Figure 6-17 on page 6-19).

Set the CENTRIFUGE SPEED control to 0 rpm. This ensures that the
centrifuge will not spin when the START SPIN button is pressed.

Press the START SPIN button, then immediately press the SUPER OUT
button. This inflates the flexible diaphragm under the cell processing bag and
expresses cell product from the cell processing bag back into the cell product
bag.

Transfer as much fluid as possible out of the cell processing bag back into the
cell product bag.

Press the STOP RESET button.

Press the BLOOD IN button to open valve V1. Replace the red-striped tubing
into valve V1, and press the STOP RESET button.

Remove the clamp from the purple-striped tubing.

Open the centrifuge cover interlock latch (if applicable), open the sliding
covers, and remove the centrifuge bowl cover and alignment blocks.
