SOP-10: Laser Frequency Calibration of the
RamanFlex 400
Laser frequency calibration should only be performed if the instrument has failed a
wavelength calibration verification.
1.
Ensure the RamanFlex 400 is switched on by following SOP-1.
2.
Click Spectral Preview in the Scan and Instrument Setup dialog, and focus on a
sample of polystyrene (L1181228).
When the polystyrene sample is optimally focused, close the Scan and Instrument Setup
dialog by clicking Exit.
NOTE: If you have a RamanFlex 400F with two fiber optic probes, ensure that the probe not
in use is covered.

Select Validate
and then Instrument from the Instrument menu in Spectrum.
The Validation / Calibration dialog is displayed.

Select the Calibration tab.

Select Laser Frequency, and then click OK.
The instrument will now perform the laser frequency calibration routine. This will take a
few minutes.
After completing the laser frequency calibration routine, a report is generated on-screen
that gives details on the outcomes of the laser frequency calibration.
In the unlikely event that the laser frequency calibration routine should fail, please
perform a wavelength calibration and repeat the laser frequency calibration.
The instrument in now laser frequency calibrated.
SOP-11: Intensity Calibration of the RamanFlex 400
The intensity calibration must be performed using the National Institute of Standards and
Technology (NIST) Standard Reference Material. NIST SRM 2241 is the relative intensity
correction standard for Raman spectroscopy when using
785 nm excitation.
Intensity calibration should typically be performed on an annual basis, that is during an
annual instrument service, or whenever the instrument is moved, or following a service
repair.
NOTE: As intensity calibration is not required on a regular basis, NIST Standards are not
included in the basic RamanFlex 400 package. Standard Reference Materials can be
purchased from PerkinElmer (L1321831). Your instrument will be intensity calibrated
prior to shipping or during installation.
NOTE: If you have a RamanFlex 400F with two fiber optic probes, you must perform an
intensity calibration on both fiber optic probes.
