Using the RamanFlex 400 . 77

SOP-6: Sample Positioning when using a Fiber Optic
Probe

You can use the Spectral Preview button to position your sample for optimum spectral

collection efficiency. Sample positioning and alignment in preview mode allows you to

position and focus on a specific area and to collect spectral data from that location. It can

also be used to enhance the signal-to-noise ratio of spectral data. The data that is displayed

on screen during a spectral preview cannot be saved.

Ensure that the RamanFlex 400 is switched on.
If not, follow SOP-1.

Select Setup

from the Instrument menu in Spectrum.

The Scan and Instrument Setup dialog is displayed.

Click Spectral Preview.
Real time spectral acquisition will commence and spectra are displayed in the spectral

preview tab.

Set the Preview Scan time to 1 second, and then press Enter.
This will automatically acquire another background. You can enter a Preview Scan time

in the range 0.1 – 10 seconds.
The sample may now be positioned and aligned correctly for optimized data collection.

The spectral data can be optimized by adjusting the distance between the fiber optic

probe and the sample.

Use the Pause and Play buttons to examine the spectrum in the Spectral Preview panel.
When you click Play the preview acquisition resumes.

You can now collect and save spectral data.
