View the specimen.
To observe another part of the sample, rotate the stage
knobs to find the position to observe. If the sample is not
in focus, rotate the focus knobs to adjust the focus.
To observe under a different magnification, repeat steps
15 and later with another objective.
Adjusting the contrast
To adjust the contrast, loosen the polarizer rotation
clamp screw and rotate the polarizer.
Adjusting the background of the field of view to a gray
sensitive color will provide high contrast for
microscopy.
Contrast is applied in the shearing direction (see the
diagram). By using a rotatable ceramic coated stage,
you can rotate the stage to adjust the orientation of
the specimen, thereby aligning the section you wish
to enhance the contrast with the shearing direction.
Microscopy with color contrast
Placing the NCB filter into the optical path and
inserting the D-LP lambda plate into the lambda plate
slot on the DIC nosepiece will turn the background of
the field of view to a sensitive color, enabling
observations with high color contrast. An interference
color is displayed based on changes in the refraction
index and/or thickness of the specimen.

When the epi-fluorescence cube turret is attached to
the microscope, the optical element for the
epi-fluorescence microscopy installed in the
epi-fluorescence cube turret is not used in differential
interference contrast microscopy. Be sure to bring
Address 1 (empty) on the turret into the optical path.
When the FL/DIC analyzer slider is inserted in the
epi-fluorescence cube turret, pull it out to the first
click position to remove the analyzer from the optical
path. When a dummy slider instead of the analyzer
slider is inserted, do not fully insert it. Pull it out one
step. If it is fully inserted, the periphery of field of view
may be dark.
