Lifetime mapping
Introduction
In this mode, the sample is moved across the objective and TCSPC measurements are
taken from pre-defined locations on the sample.
DataStation allows you to define the set-up parameters, specify the measurement loca-
tions on the sample, run the measurement scan, and eventually display contour maps of
the results. Subsequent analysis can be performed in the DAS6 decay analysis package.
We now assume that you are familiar with the single-point measurement routines de-
scribed earlier. For best results we advise you to perform at least one single-point
measurement on a sample before running a scanning-mode experiment.
Two mapping modes are offered:
TCSPC multi-point
mapping
Acquisition time is larger than 1 s per decay. It is possible to
map non-contiguous non-rectangular areas.
TCSPC FLIM Acquisition time ranges from 5 ms to 1 s per decay. Only rectan-
gular areas can be mapped.
Anchor control
An additional anchor control is available in this mode. The anchor control allows you
to select the area of the sample that is going to be scanned. The use of this control is de-
scribed in the measurement example later in this chapter.
Pre-measurement procedure

Before starting scanning mode, follow the
measurement instructions in single-point mode
to make sure that suitable data can be obtained
from the sample.
Ensure that the set-up conditions are correct using the single point manual mode
before going into the more-automated and less-flexible scanning mode.

When you are satisfied with the experimental
conditions, start a New Measurement in the File
menu and select the TCSPC
– Multi point map-
ping icon in the New Measurement window.
Note:
If the system is running in reverse mode, choose this op-
tion while in Single-point mode.
