Objective
In order to get an accurate positioning of the microscope stage,
the objective value should always reflect the magnification of the
objective that is currently used on the microscope. This value can
be changed in the
Microscope Objective
window (Fig. 11):

Calibration procedure
The calibration procedure teaches the system the relationship between the positions of
the microscope motorized stage and the camera pixels, and lets the system know where
the pinhole is located in the horizontal plane. Therefore, run the calibration routine each
time that the system configuration changes; for instance, when the pinhole location has
been modified, when the camera orientation has changed, or when the mechanical stage
configuration has been modified. During the calibration process, make sure that the ob-
jective value reflects the objective currently being used on the microscope.
There are two types of calibration:
Calibration of stage and camera relative position, also called Stage calibration
Calibration of laser-spot position and diameter (if applicable), also called Video cal-
ibration
Note:
Time-resolved measurements are still possible when the illumi-
nator is in position

, but the excitation efficiency is halved and then
there is no discrimination of the laser.
