Calculation based on partial images:
The image detected through the disk contains mainly intensities from the focal plane, however
superimposed with a widefield portion. In order to obtain an optical section, this widefield intensity must
be subtracted. For this purpose, the system uses the second, simultaneously detected image, which was
reflected from the disk and thus contains "out-of-focus" intensities. An optical section is obtained by
simple scaled subtraction of both images:

In this case,
K
represents a fixed constant correcting differences in the light efficiency of both imaging
pathways.
A scaled addition of both images yields a conventional widefield image. Thus, all the information from
the specimen is captured by VivaTome in a raw data image and is available for further analysis.
