A warning popup will appear if the user launches a run starting with the
“Partition”
command of the Sapphire chip (resp. Opal chip) and ends with the
“Release” command
of the Opal chip (resp. Sapphire chip), and if the user launches a run starting with the
“Pressurization” command.
•
The internal source pressure of the Geode during the cycling step is fixed by Stilla
Technologies and should not be modified by the user for optimal results. If a modification
is required, please contact Technical Support before proceeding
.
Run
1)
In the main panel, press “Run
”, then press the “open icon”
. In the “Open program”
window, select the program using the drop-down menu
in the “local/scripts” or “/templates” sub-
folder and press the
“open icon”
.
2)
Optional (not recommended): in the main panel, press “Run
”, then “temperature ” to fix
the temperature of the thermal plate without time limitation. Do not exceed 25°C for more than
20 min before starting a run.
3) Close the lid and then press the
“play icon”
. The remaining time (indicative) is displayed,
as well as the progression bar of each step (based on the remaining time), the current measured
temperature of the thermal plate, and the current measured pressure in the block (
Fig. 11
). Press
the
“information icon”
to get a summary of the program.

4) Visualize the temperature profile of the thermal plate and the pressure profile in the Geode
chamber during a Crystal Digital PCR
TM
(automatically recorded) by pressing on “Live Logs” in
the main panel.
Note: Only the “Live Logs” of the most recent experiment are automatically saved in the Geode.
The temperature profile is displayed in red color and the pressure in blue color (
Fig. 12
). Use the
arrows and magnifying buttons to navigate through the diagram. The live record will be finished
as soon as the PCR program has ended. It can be exported by plugging a USB key and pressing
the “export icon”
