90 . Spotlight 150 User's Guide

Lower the stage and click

The automated ATR crystal will be lowered automatically. The spectrometer starts to

scan and the background spectrum is displayed as it is collected.

Collecting a Sample Spectrum

Once you have collected a background spectrum, follow the procedure below to collect a

spectrum of your sample.

Make sure that the ATR crystal is retracted.

Place the sample slide on the stage.

Move the sample to the center of the field of view.
Even though the microscope may not be focused on the sample, you can see a change

in the light intensity as the sample moves into the center of the field of view.

Look at the image on the Setup Microscope tab and focus on the sample using the

microscope focus controls, then move the area of interest into the center of the field of

view.

Select

Monitor

from the Measurement menu.

The Live tab is displayed. The software will prompt you to lower the stage to prevent a

collision when the ATR crystal is lowered.

Lower the stage and click

The automated ATR crystal will be lowered automatically.

NOTE:

Before you collect the sample spectrum, you should monitor the spectrum to make

sure that there is good contact between the crystal and the sample.

Raise the stage carefully until the sample touches the crystal.

CAUTION

To prevent accidental damage to the ATR crystal, always bring the

sample slowly up to the crystal, rather than lowering the crystal directly

on to the sample.

Select

Sample

The spectrum at the current aperture position is displayed.

Make sure that the spectrum monitored is satisfactory.
If your sample is soft or there is poor contact, you may need to increase the contact of

the crystal with the sample, by raising the stage slightly.

When the spectrum is satisfactory, click

Scan

The software may prompt you to raise the stage to make contact with the sample. Click

to proceed. The spectrometer starts to scan and the spectrum is displayed as it is

collected.
