6. Instrument setup– InspectIng the phase contrast rIng
6.2 Inspecting the phase contrast ring
For the Leica DMi8 with tiltable transmitted light illumina-
tor arm
If your microscope is equipped for the use of phase con-
trast, the light rings that fit the objectives are built into the
condenser. The light rings are already centered in the fac-
tory. As a result of transport and setup of the stand, how-
ever, in some cases centering maybe be required again.
Therefore check the centering.
Note:
Every objective is assigned its own light ring in the con-
denser disk. Therefore, a check must be performed for
each objective.
Regular phase contrast with phase objectives
When rotating in an objective that is suitable for phase con-
trast, the corresponding light ring is introduced automati-
cally (motorized condenser) or you must manually swing
the corresponding light ring inwards.
Leica DMi8 with manual condenser:
• If necessary, set the TL bright-field position or empty po-
sition on the fluo turret, or select the "DAPI-TL" filter cube.
• Select the bright-field position on the condenser.
Leica DMi8 with motorized condenser:
• Select the
PH
contrast method (phase contrast).
• Instead of an eyepiece, insert the focusing telescope
(Fig. 53) into the observation tube.
• Select the phase contrast objective with the lowest mag-
nification.
• Optimize the focus on the specimen.
• Put the ring structure (Fig. 55a) by into focus by slightly
loosening the clamping ring (Fig. 53.2) and moving the eye
lens (Fig. 53.1).
• Retighten the clamping ring.
