This chapter describes how to perform the following procedures:

• Set up the Ion Chef

Instrument for use by diluting the libraries, and loading the instrument with all

of the required reagents and consumables

• Start an Ion Chef

 run

• Unload the chips for sequencing

Materials required

• Ion 520

 & Ion 530

ExT Kit –  Chef (Cat. No. A30670)

• Ion 520

 Chip or Ion 530

 Chip

• Library or combined library stock solution
• Molecular-biology grade nuclease-free water
• P200 pipettor and filtered tips
• Waste container

Dilute the library

Sample libraries are quantified at the end of the library preparation workflow and are typically diluted
to a concentration of 100 pM. The Ion Library Equalizer

 Kit procedure also normalizes library

concentration to 100 pM. Libraries may require further dilution before use in Ion Chef

 template runs.
