Action
Step
Make sure that the mobile phase top and mobile phase bottom inlets are
closed, then connect the packing pump to the hydraulic inlet (HI) and close
the hydraulic outlet (HO).
1
Open the bottom slurry valve (BSV) to allow pumping to the slurry tank (A).
2
Start the packing pump at a rate of around 50%. The hydraulic chamber will
fill and the adapter will move downwards as the slurry is removed through
the bottom slurry valve.
3
When pressure increases in the hydraulic chamber (the adapter is moving
against a packed or sedimented bed) stop the pump.
4
Use the slurry to continue the unpacking of the column. This avoids the need
to use large volumes of unpacking buffer and larger buffer tanks.
5
Open the hydraulic outlet.
6
Connect the slurry tank to the filling pump (membrane pump) to the bottom
slurry valve. Keep the hydraulic inlet (HI) and the mobile phase top and mobile
phase bottom openings closed.
7
Pump slurry into the column to fill it with 1 CV.
8
Repeat steps 7 to 13 until the adapter is able to reach the lowest position.
9
Flushing the column
