Troubleshooting
AM-190 Manifold
Problem
Possible Cause
Possible Solution
Gas leaks from the safety
relief valve.
1. Gas supply pressure is too high.
2. Safety relief valve is defective.
3. Safety relief valve is not properly
installed.
1. Set the inert gas supply pressure to between 100 and 125 psi
(6.9 to 8.6 bar).
2. Order a new safety relief valve (P/N 02196).
3. Remove and re-apply PTFE tape, and then re-install using the
appropriate wrench.
Inert gas tubing blows/pulls
free.
1. Tubing is damaged.
2. Instant-tight fitting is defective.
1. Cut a new end on the tubing, re-insert, and repeat the “pull test”.
2. Replace the instant-tight fitting (P/N 02184).
Pressure gauge is stuck.
Pressure gauge is defective.
Replace the pressure gauge (P/N 07900).
Water in the inert gas tubing
or the solvent reservoir.
Water trap is full.
Drain the water trap; place a waste container underneath the trap and
loosen the screw at the bottom of the trap (see Figure 4 on page 3).
Check the inert gas supply for excessive moisture content.
Gas leaks in the AM-190
manifold.
Modular connector screws have
loosened.

Flash 75/150 Radial Compression Module
Problem
Possible Cause
Possible Solution
Gas leaks from top or bottom
of the radial compression
module.
1. V-band clamp is loose.
2. O-ring seal is leaking.
1. Tighten the upper V-band clamp. Sometimes it helps to loosen the
lower V-band clamp just a bit before tightening the upper, and then
re-tighten the lower V-band clamp.
2. Remove the head assembly and replace the O-ring (P/N 00778 for
Flash 75 and P/N 01420 for Flash 150).
Gas leaks from the safety
relief valve.
1. Radial compression pressure is
too high.
2. Safety relief valve is defective.
3. Safety relief valve is not properly
installed.
1. Set the radial compression pressure to between 80 and 100 psi
(5.5 to 6.9 bar).
2. Replace the safety relief valve (P/N 00790 for Flash 75 and
P/N 02196 for Flash 150).
3. Remove and re-apply PTFE tape, and then re-install using the
appropriate wrench.
Top head assembly will not
seat.
1. Bottom head assembly is out of
alignment.
2. The cartridge is too long.
1. Loosen the bottom head assembly, re-align to center, and tighten
the V-band clamp.

Gas bubbles in the collection
line.
1. Solvent reservoir is empty.
2. Knife edge seal leaks.
1. Refill the solvent reservoir as described on page 18.
2. Tighten the upper and lower V-band clamps.
Solvent leaks inside of the
barrel.
1. Radial compression pressure is
too low.
2. Solvent pressure is too high.
3. Knife edge seal leaks.
1. Set the radial compression pressure to between 80 and 100 psi
(5.5 to 6.9 bar).
2. Ensure that the solvent pressure is at least 20 psi (1.4 bar) lower than
the radial compression pressure.
3. Remove the cartridge and check the top and bottom lip for full
penetration. Install a new cartridge if necessary.
Flow runs backward toward
the SIM.
Solvent pressure is higher in the
cartridge than in the SIM.
Always wait until all flow stops from the fraction collection line before
opening the three-way valve on the radial compression module toward
the SIM.
Inert gas tubing blows/pulls

1. Tubing is damaged.
2. Radial compression pressure is
too high.
3. Instant-tight fitting is damaged.
1. Cut a new end on the tubing, re-insert, and repeat the “pull test”.
2. Set the radial compression pressure to between 80 and 100 psi
(5.5 to 6.9 bar).
3. Replace the instant-tight fitting (P/N 02184).
Troubleshooting
Biotage Flash systems are robust and operate with a minimum
amount of required maintenance. This section explains how
to troubleshoot some commonly occurring situations where
the system, or one of its components, does not seem to be
working properly. The most common occurrences are simple gas
leaks. To locate the source of a leak, we recommend that you
apply a leak detection liquid or soapy water to the suspected
area. Once the leak has been located, adjustments or repairs
are relatively straightforward. However, if any assistance is
