7. Advanced Settings for Melt Curve Analysis:
• Analysis by Melt Curve will need to switched ON using the toggle switch for that option
. The background behind the switch will turn blue when activated.
• Tap the settings icon to access advance setting for the Melt Curve.
• The starting and ending temperatures can be adjusted if the default options are not
acceptable. The default values for the melt curve are from 60–95ºC.
• The resolution of the melt curve can also be adjusted. There are three different levels of
resolutions available. The default level of resolution is set at ºC. When set at 1ºC, this
means for every increase of temperature of 1ºC, the plate will be scanned, and data will
be recorded. Higher resolution means more scans are performed, acquiring more data
with each scan. 0.1 ºC is the highest resolution available with the Cielo Real-Time PCR
system. To save any changes made to the Melt Curve settings, select MODIFY
at
the bottom of the window. To close out of the menu without retaining the changes, select
CANCEL
.
• This will open a new window where an alternate ramp rate between 0.1 and 6.0º C
per second can be entered. Once the desired value has been entered, hit DONE to
return to the Advanced Settings Screen. Finally, hit MODIFY to retain the entered ramp
rate or CANCEL to abort the new setting.
