RamanMicro 200 Series Getting Started Guide
SOP-4: Positioning the Sample on the Microscope
1.
Lower the microscope sample stage.
2.
Place the sample on a microscope slide and place it on the stage.

Select the 50x objective lens on the microscope.
4.
Move the sample so that it is approximately lined up below the objective lens.
5.
Select the Bright Field (BF) switch position on the Olympus illuminator module.
6.
Switch on the illumination source.
7.
Open the Scan and Instrument Setup dialog, and on the Hardware tab select
Microscope 50x from the Beam path drop-down list.
8.
Focus on the sample using the video camera.
Use Video Preview on the Scan and Instrument Setup
dialog. Take care not to crush
the sample.
9.
Use the stage to accurately position the sample in the x and y directions.
10.
If higher magnification is required, select the next most powerful objective lens on the
microscope.
The microscope is supplied with 5x, 20x and 50x objective lenses.
NOTE: Do not touch the glass of the objective lenses when you select the appropriate
magnification.
11.
On the Hardware tab select the appropriate objective lens from the Beam path drop-
down list.
12.
Focus the sample and again move the sample in x and y directions.
The crosshair indicates the spot where analysis takes place.
13.
When the sample is correctly positioned, switch off the illumination source.
