E.2. Gap between sample and medium
The filling of the mould with medium must be done immediately after the sample transfer. If not, the sample
starts to freezing and doesn’t link with the medium. The gap between sample and medium make the cut
difficult.
Solution
Trim the block until the gap is gone away.
How to avoid
1. Fill the mould with medium immediately after the sample is transferred inside the mould.
2. Make sure to have an available bottle with enough medium to fill the mould at the right moment.
3. Prefer Milestone Cryoembedding Compound (MCC) which, being more fluid, adheres better to the
sample.
E.3. Block sticking to the mould
At the end of freezing, it may
happen that the block doesn’t exit from the mould. This happen when there is
frost inside the mould that act as a glue between mould and MCC block.
Solution
Vertically hit the chuck with the heat extractor. This will break the link between block and mould and make
the block free. Don’t use the tweezers like lever.
If the removing it is very difficult, unscrew the mould and put it under cold water for very few seconds, in
order to melt the linking layer and detach the frozen block.
