12) Remove the frozen specimen and chuck when timer alarms. It will be flat and the surface will require
less trimming and wastage of tissue before the entire tissue face is available for sectioning.
3.10.1. Freezing time
The freezing time depends on the size of the mould used and on the sample type.
The mould for 30 mm chuck provides a standard freezing time of 40 seconds. In case of delicate specimens
which are cut in cryostat at temperatures equal to or greater than -20°C, the freezing time can be reduced up
to 20 seconds, so as to have a less hard block and easier to cut. In case of fatty tissues we recommend
extending the freezing time to up to 80 seconds, so as to have a harder block that helps to cut the adipose
tissue.
The mould for 40 mm chuck provides a standard freezing time of 60 seconds. In case of delicate specimens
which are cut in cryostat at temperatures equal to or greater than -20°C, the freezing time can be reduced up
to 30 seconds, so as to have a less hard block and softer to cut. In case of fatty tissues we recommend
extending the freezing time to up to 120 seconds, so as to have a harder block that helps to cut the adipose
tissue.
3.10.2. Using multiple timers
The “multiple timers” button on the Chiller page opens a screen with 6 timers:
More than one timer can be useful in some cases:
-When it is necessary to freeze more samples simultaneously.
-When different size moulds are present in the PrestoCHILL.
-When wanting to freeze different tissues in the PrestoCHILL with specific times.
