Step 3 – Set the Acquisition Protocol
• Switch to the Protocol Tab
• In the Prepare box, enable a 30 second pre-plate prime
and a 30 second pre-plate shake.
Note that other prime and shake patterns can be used. If
you suspect insufficient sample resuspension increase the
duration of the pre-plate shake. In total, however, it is best
to have 60 seconds of combined pre-plate prime and shake
(prime + shake = 60 seconds).
• In the Sample box, set the Sip Time to 2 seconds and
choose a mapped 96 well plate model that matches the plate
you prepared.
IntelliCyt recommends collecting 1000–2000 events of
interest per well for statistical relevance in most assays.
IntelliCyt also recommends the Standard (29 RPM) speed for
most assays. At this speed, the instrument delivers between
1.5 and 2 µL per second. Taking your cell concentration and
the percentage of cells that qualify as events of interest (for
example, CD4+ normal singlets), you can calculate Sip Time.
In this example:
• Cell concentration = 1000/µL.
• Events of interest (normal singlets) = ~65% of the initial cells
• 2 second Sip = ~3.5 µL = ~2,275 normal singlets/well.
