Regular cleaning
Regularly wipe the outside of the column using a cloth moist with an antibacterial agent.
This reduces the risk of microbial growth.
General method for
sanitization-in-place

It is recommended to rinse the column with purified water, 2 column volumes upflow
at a flow velocity of 60 cm/h

Flush the column with 1.0 M NaOH:
a
2 column volumes upflow at a velocity of 60 cm/h
b
2 column volumes downflow at a velocity of 60 cm/h

For 50 and 70 columns with plastic bed support: Recirculate 5 column volumes of
1.0 M NaOH, upflow for 2 hours followed by downflow for 2 hours at a pressure of
1 bar.
For other column configurations: Recirculate 5 column volumes of 1.0 M NaOH, upflow
for 4 hours at a velocity of 60 cm/h.

Wash the column with buffer or water of desired quality to achieve a neutral pH.

Re-equilibrate the column with storage or starting buffer.
