Action Step
Storing: After imaging, the gels can be stored in the dark at -8°C
for several weeks in storage solution (pH 2.3).
Note:
This allows for further imaging at a later date, if required. For long
term storage (up to 6 months) add Deep Purple stain in 1:200
dilution.

Silver staining
Introduction
This section presents a Silver staining protocol recommended by GE
Healthcare. Refer to Silver Staining Kit Protein 71-7177-00 for more
information.
Silver staining solutions
Preparations Solutions
1
Mix 75 ml ethanol and 25 ml glacial acetic acid.

Make up to 250 ml with distilled water.
Fixing
solution
1
Mix 75 ml ethanol, 10 ml sodium thiosulfate (5% w/v)
and 1 packet of sodium acetate (17 g).

Make up to to 250 ml with distilled water.
Before use: Add 1.25 ml glutaraldehyde (25% w/v).
Sensitizing
solution
1
Mix 25 ml of silver nitrate solution (2.5% w/v) with
distilled water.

Make up to 250 ml with distilled water.
Before use: Add 0.1 ml formaldehyde (37% w/v).
