Method 1: Positive Pressure
1. Unclamp the pumps by pressing upwards at the dotted triangle on the
pump head.
2. Place the tubing onto the pump rollers. The colored tubing marker should
be to the left of each pump and match the colored flag next to each pump
(Figure 2).
3. Leave the pumps unclamped.
4. Connect a pipettor to the sterile filter of the inlet bottle and use the “pipette
down” button to pressurize the inlet bottle (Figure 3). Cell solution will flow
from the seeding bottle, through the tubing, and into the flasks.
5. Tilt the seeding bottle to allow all the liquid to flow into the dip tubes. This
ensures that all of the cell solution is perfused into the flasks.
6. Stop pipetting when the seeding bottle is empty. The inlet side tubing will
still be filled with cell solution.
7. If cell solution enters the outlet tubing, “pipet up” to reverse flow.
8. Clamp the tubing onto the pumps by pressing down on the pump head clamp.
9. The tubing holder on each side of the display may be used to help secure the
tubing in place.
NOTE:
Corning® MicroDEN® flasks can be filled separately by clamping one of
the tubes in the pump to stop flow. Liquid will flow through the unclamped tube.
Method 2: Pump Perfusion
1. Unclamp the pumps by pressing upwards at the dotted triangle on the pump
head.
2. Place the tubing onto the pump rollers. The colored tubing marker should
be to the left of each pump and match the colored flag next to each pump
(Figure 2).
3. Clamp the tubing onto the pumps by pressing down on the pump head
clamp.
4. Turn the Corning® MicroDEN® unit “ON,” and select “Seed” using the function
button on the unit.
5. The pumps will perfuse the cell solution from the seeding bottle into the flasks.
6. Tilt the seeding bottle to allow all the liquid to aspirate into the dip tubes.
This ensures that all of the cell solution is perfused into the flasks.
7. Stop perfusion by pressing the MicroDEN
unit function button when the
seeding bottle is empty. The inlet side tubing will still be filled with cell
solution.
NOTE:
Corning MicroDEN flasks can be filled separately by unclamping one of
the tubes to stop flow. Liquid will flow through the clamped tube.
Helpful Seeding Tips

During seeding, tilt the inlet bottle to allow all the liquid to flow into the dip
