1. Press the
Confluence menu
.
2. Select one image (Bright image) file whose confluence value
should be recalculated.
3. Adjust the image by changing parameters.
- Refer to the recommended sensitivity and background corresponding to cell line.
4. Press the
Confluence button
. The confluence value would
be recalculated by new sensitivity level and background level.
· If the confluence value from step 4 is good enough to use, then check the
Apply
box
to apply new sensitivity and background level to the rest of images in the same
folder.
Check the channels
(Channel 1 or Channel 2)
and press the
OK button
to get
new confluence value.
5. Confluence graph would be redrawn.
6. Press the
Save as button
to save the images with the new
confluence values.
