Then the equation for finding the concentration of the measured ion is
This is the algorithm with which the 103APV4R DIESTRO Analyzer works.
Principle of operation of TCO2 determination
The determination of TCO2 is carried out as follows:
A similar amount of Lactic Acid (L.A.) is added to a fixed amount of serum (
140
uL
)
that decomposes all
the bicarbonate found in the sample according to the equation:
C
3
H
6
O
3
+ NaHCO
3
→ CO

+
H
2
O
+ NaC

lactic acid + sodium bicarbonate --> Carbon Dioxide (gas) + Water + Sodium Lactate
The reaction is aided by a magnetic stirrer that mixes the L.A. and the sample in a constant volume
reactor for a certain time.
The gaseous CO
2
released in the reactor produces an increase in pressure that is measured with a
sensor connected to it. This pressure increase is directly proportional to the amount of NaHCO
3
in the
sample.
The equipment is previously calibrated with a solution with a known concentration of NaHCO
3
, with the
pressure increase measured, a calibration constant is determined and then applied for the determination
of the TCO2 of the sample. In both cases, temperatures before and after the reaction are also measured
to compensate for the increase in temperature pressure.
