Preparatory Steps

For the basic adjustment of the stand and essential explanations, please see the current operating

manual of the Laser TIRF imaging system.

Screw the two adjustment tools included in the TIRF adjustment tool kit onto the nosepiece of the
Axio Observer, preferably beside the TIRF objective. If necessary, use the M 27x0.75 adapter on

W 0.8, H = 0.

Insert the reflector module equipped with neutral-density filters into the reflector turret.

When using motorized instruments, make sure that there is no focus offset between the TIRF
objective and the adjustment tools.

Prepare a test specimen with fluorescing beads, possibly by means of the TIRF specimen kit (433800-

0000-000).

Focus on the interface from cover slip to medium, that is, on adherent fluorescing beads in the dish

or on the cover slip.

See the table below for use of the adapter rings.
