Chapter 3 Freezing Samples
Starting the Run
Never leave the Kryo550-16 unattended while running a profile.
Make sure that there is sufficient liquid nitrogen to complete the freezing profile.
If using a EuroCyl or CryoCyl cylinder make sure the supply tap is open.
If using a Planer LNP4 pump and dewar, switch on the pump power supply and wait for
the orange light on the pump power supply to go off; this indicates that the correct
pressure has been reached.
It is normal to load the samples after the chamber has reached the start temperature.
If you intend to run from a PC, initiate the profile from the PC using the Planer plc
application -
DeltaT . Refer to the DeltaT documentation for details.
If using the kryo-connect module, refer to the kryo-connect documentation for details on
running a profile.
If you want to run a profile directly from the control panel, you must have
downloaded a valid profile from
DeltaT first into the Kryo550-16's
Permanent Profile
memory. See

If running directly from the Kryo550-16, just press the Run button on the control panel.
The chamber will start to be heated or cooled until the start temperature is reached.
When the start temperature has been reached , the Run Indicator on the control panel will
start to flash and the Kryo550-16 will begin to beep.
Loading the Samples
Always use tongs or wear cryogloves when handling cold objects.
Load the samples. This will depend on the freezer furniture fitted to your chamber. Note
that during this time the beeping can be cancelled, without starting the run by pressing
the Sample key.
After loading all of the samples, it is advisable to wait five minutes to allow the
Kryo550-16 to re-stabilise before pressing the Run button on the control panel. The beep
should stop and the Run indicator should remain illuminated but stop flashing.
If manual seeding has been selected for the profile the Kryo550-16 will beep again after
the defined soak period and the display will show S, indicating that the samples are ready
for seeding. See

After seeding, press Run again to continue the profile.
At the end of the run , the freezer will again prompt you by beeping and flashing the Run
indicator when the samples are ready for removal. The Kryo550-16 holds the chamber at
the end temperature until you confirm that all samples have been removed ; see
