Before each wash procedure and each row the program checks whether there is
an electrical connection between the dispensing and aspirating needle(s). If a
connection exists, the dispense and aspiration errors can not be detected and the
program is interrupted immediately or will not even be started
The underside of the manifold must then be cleaned, as the deposits from the
wash buffer can produce an electrical connection between the needles, which
could prevent the Fill Verification option from functioning correctly.
Test Conditions
The following conditions are required for the Fill Verification to function flawlessly.
•
The conductance of the wash buffer must be between
5 mS/cm
and
30 mS/cm
.
•
The amount of wash liquid dispensed must produce at least
100 µl overflow
.
•
At least
450 µl
should be dispensed with an Overflow Position slightly above
the upper well limit.
Conductance of some Standard Wash Buffers
Most of the commercial wash buffers lie in the mentioned conductance ranges.
The conductance is proportional to the wash buffer concentration.
The following values were measured using a normal dilution:
