4.2 Unpacking the column
Step Action
1 Disconnect column from the system

Position a beaker or connect a piece of tubing from the column outlet to a
flask to collect the acetonitrile that will exit the column.

Apply a low compressed air flow at the inlet of the column to allow the
Acetonitrile to come out at the bottom of the column.

Continue the air flow until the support has reduced in volume and is free
from the column wall. This will take approximately 30 minutes depending
on the amount of support in the column and the air flow.
5 Turn off the air flow

Disconnect the top end piece by turning the bayonet anti clockwise and
lift it up
7
Turn the column upside down and collect the support in a suitable vessel.
8 Start the deprotection as described in the ÄKTA oligopilot User Manual.
