Spectrophotometer Principle and Intended Use
UV/Visible spectrophotometers measure the transmission of light through a sample. Samples absorb light
based on their unique molecular composition. The amount of absorbance is directly proportional to the sample
concentration and the pathlength, which is the distance that the light travels through the sample.
The WPA Lightwave spectrophotometers are intended for laboratory use to measure the concentration of nucleic
acids and proteins in experimental samples.
Hardware
Your spectrophotometer is a simple-to-use UV/Visible instrument with a CMOS array detector (1024 pixels).
It has no moving parts, which is the basis of the rapid scanning operating system. The look and operation
of the Lightwave 3 and 3+ are identical; the only difference between them is the bandwidth. Throughout the
rest of this manual, the term Lightwave 3 will be used to cover both instruments.
