1 Setting up a Run for Purification of Genomic DNA
from Whole Blood Samples
This procedure describes the purification of genomic DNA from 96 samples of 200 µl fresh or frozen
whole blood using the MagNA Pure 96 DNA and Viral NA Small Volume Kit. Setting up a run for puri-
fication of genomic DNA comprises the following steps:
1. Preparing the samples including internal control, see next section.
2. Defining the parameters in the software including internal control and sample transfer, see section
Defining the Parameters in the Software .
3. Loading the instrument, see section Loading the Stage .
1.1 Preparing the Samples
Always use sterile, nuclease-free pipettes.
To prepare the internal control
Commonly used internal controls (ICs) monitor the nucleic acid isolation efficiency.
Remove the cover from the MagNA Pure 96 Internal Control Tube and pipette 3100
µ
l of the internal
control reagent into it.
The volume of the internal control is given for MagNA Pure 96 Software version 2.0. The respective
volume is different when you use the software versions 1.0 or 1.1. For detailed information see the
Instructions for Use of the respective kit.
Close the MagNA Pure 96 Internal Control Tube.
In the MagNA Pure 96 Software, click the Settings button in the global action bar. The Settings dialog
box opens.
