Packing procedure
To pack the column follow these steps:
Step Action
1 Add the required weight of dry Primer Support to a beaker. See Calculate
column volume, on page 22 for information about required weight.
2 Make a slurry by adding 1.2 CV of acetonitrile to the beaker, stir gently and
allow Primer Support to swell for 2 to 3 minutes.
3 Remove the adapter from the column. See Section 6.1 Disassemble and
assemble for cleaning and maintenance, on page 26 for more information.
4 Add the slurry to the column. Allow the Primer Support to settle for 2 to 3
minutes.
5 Insert the adapter. See Section 6.1 Disassemble and assemble for cleaning
and maintenance, on page 26 for more information.
6 Adjust the column adapter to the level that corresponds to the calculated
volume from the list in Calculate column volume, on page 22.
7 Connect the system to the inlet of the column using a PTFE tube with an M6
connector (18117264) at the column side and a connector (18112117) at the
system side, using a ferrule on each end.
Tip:
Keep the tubing as short as possible.
Note:
Other adapters from 5/16" to M6 can be used for the inlet. It can be
necessary to lift the column, since other adapters might not fit between the
column and the bench surface.
8 Connect the system to the outlet of the column, using a tube with any
connector and a ferrule on each end.
9 Start the UNICORN™ method instruction
PFlow AB (10 bar flow) on the
connected system. Continue for 1.5 CV to 2.0 CV.
10 Stop the flow.
11 Start the run.
