Once the Live Image resumes (Camera ON), the user will see the Live feed of the IrysChip concentration zone
(blue gradient at bottom border of Live Image screen) and leakage zone (blue gradient entering remaining portion
of the Live Image screen). The blue gradient is the Live Image of the laser-illuminated DNA backbone; this laser-
illumination is at a low exposure to minimize photo-degradation.
Loading Step: Concentration
Once the user is able to visualize the DNA (horizontal blue band at the bottom of the Live feed window), Irys
guides the user through Concentration optimization. Irys asks the user to press the OK button each time they see
a vertical stream of DNA entering the leakage zone. Pressing OK will decrease the force, which stops the stream.
There is a five-second period after each force decrease during which the OK button is disabled.

Loading Step: Spike
Once the user completes the Concentration steps, Irys moves to the next loading step, Spike, where the DNA that
is concentrated at the front (horizontal blue band indicates DNA is gathered at edge of the Inlet) is loaded into the
Flowcell. The Live image becomes inactive, displaying Camera OFF.
Note:
Because the Spike occurs quickly, there is no timer for this step.
