Acquire an image (transmission/fluorescence) with the black/white camera
Before observing in brightfield, you have to set the
Khöller illumination, then if you need to the phase
contrast or DIC (refer to the posters in the microscope
room).
Select « BF », « PH » ou « DIC » on the TFT screen.
1.
Select the tab « Acquisition ».
2.
Choose « Fluo + TL - Camera N&B ».
3.
Open the menu « Channels ».
4.
Check the line(s) with your fluorophores of interest.
5.
Select one channel (light grey).
6.
Make a « Live ».
7.
Set the exposure time automatically by clicking « Set
Exposure »
Or
8.
Set manually the exposure time.
9.
Do it for every color of interest.
10.
Make a « Snap » to image the checked channels.
