3 Fill the cuvette up to the notch (2) on the side with the selected imaging solution.
4 Place the cuvette securely on its holders again by holding the piston.
NOTICE! Be careful not to spill any imaging solution.
5 Lower the piston carefully until the bottom of the microscope chamber is reached.
6 Fasten the cover plate by tightening the screw.
7 Attach the objective lens.
5.2 Setting the parameters for the alignment
1 Start the ImSpector software.
2 Select the appropriate Instrument Mode in the Measurement Wizard window for the ordered
configuration, such as Zoom Body .
3 Select the excitation and emission wavelengths under Filter for Measurement ( Channel Setup in
software version 7.4.0 or later) in the Settings 1 window of the UltraII view. For the alignment tool, the
excitation is 561 nm. The selection must be highlighted to become active.
4 Set Laser Transmission Control . A value between 5% and 10% is recommended.
