Best Practices for Sample Enrichment and Preparation
Tips for Sample Preparation
•
After lysis, has been completed, unopened lysate can be stored at 2-8°C for up 7 days or at -20°C for up to
14 days before hydrating PCR tablets.
•
Opened lysates may be stored for up to one week for later testing when frozen at -20°C.
•
For re-testing purposes, samples can be stored after all enrichment steps have been completed, including
regrowth when required. Final enrichments (after regrowth) can be stored at room temperature until test
results have been reviewed and accepted (up to about 4-5 hours unless otherwise validated internally).
Note: If storing final enrichments for more than 5 hours, refrigerate at 2-8°C until test results have been
accepted (up to 48 hours). Storing primary enrichments before regrowth is not recommended.

For confirmation purposes, only, primary enrichments (before regrowth) could be stored at 2-8°C overnight
or according to your laboratory SOP. However, these storage conditions have not been validated by
Hygiena.
•
When preparing enrichment broth, use deionized water.
Tips for Enriching Samples
•
Unless stated otherwise, all enrichment media should be pre-warmed to the specified temperature before
use.
•
Media should be warmed in a static incubator overnight.
•
Use a thermometer to ensure incubators are within ±2°C of the specified temperature before placing sample
in the incubator for enrichment.

Tips for Transferring Samples
•
Always use proper aseptic technique, including new, powder-free gloves and new filtered or barrier pipette
tips for each step of sample preparation.
•
Break cluster tubes apart before adding sample enrichment to help prevent cross-contamination between
samples during transfer steps.
