If not already set at this value, change the
Dispensing strength
to be 1 V and the
Spraying strength
to be 12 V. These settings regulate the ultrasonic pumping action within
the dispenser while dispensing fluid and emptying the glass needle, respectively. The values
listed should be good defaults for aqueous solutions within a dispenser having a tip inside
diameter of between 10 and 50 microns. If you later find that the spot deposition on your surface
is inconsistent, try decreasing or increasing the
Dispensing strength
value. Likewise, if
your dispenser is not completely emptying itself during rinse cycles, increase the
Spraying
strength
. When done, close this window and return to the main interface.
Save the current position of the dispenser so that it will be easy to return to later. To do this,
click on the + button in the
Positions
window.
This will create a new position and leave you able to edit its name. Type in
Above slide
and
hit
Return
when you are done. The new position is added to the list of saved positions,
meaning that it will be easy to return here in the future.
It is now time to load solution into the dispenser. Place approximately 50 microliters of an
oligonucleotide solution in a well within a 384- or 1536- well microtiter plate on the deck or use a
custom well within the reach of the positioner. Manually move the dispenser above the well
(using gross positioning of 10000 micron steps, then finer positioning using 1000 micron ones)
and then down into the well (again, starting with 10000 micron steps, followed by 1000 micron
ones).
Warning
: Be careful when moving the dispenser near a solid object, such as the wells. If
the glass tip comes in contact with the side or bottom of the well, it can shatter.
