3.8.  Set up freezing parameters 

 
IN  MANUAL  MODE:  To  set  up  freezing  temperature  and  duration,  enter  in  the  Chiller  page,  through  the 
“Start chiller” icon. 
 

 

 
IN  AUTOMATIC  MODE:  To  set up  freezing  temperature  and  duration  must  be  in  the  working  chiller  period 
(pre-set in automatic cycle settings- see chapter 3.7.1) otherwise you can not access the screen below. 
 
When you are in the screen displayed below, modify the set temperature with the right arrows (red arrow to 
increase  and  blue  arrow  to  decrease)  or  by  touching  the  set  temperature  box  (in  this  case  a  keypad  will 
appear). 
Modify the set freezing time in with the lateral arrow or by touching the Time box (in this case a keypad will 
appear). 
 

 

 

 
 

3.9.  Defrost 

 
The  defrost  procedure  allows  to  obtain,  in  a  short  time,  the  complete  elimination  of  frost  that  forms  during 
normal PrestoCHILL use.  
Defrost procedure consists of plate heating, while an aspiration removes aqueous vapors derived from frost 
evaporation.  
Since samples prepared in PrestoCHILL  are fresh and so potentially dangerous, defrost vapors are filtered 
through an HEPA12 filter (blue arrow on the picture below), which removes biological pathogens.  
Then  vapors  condense  in  the  cool  trap,  positioned  in  the  left  angle  of  the  PrestoCHILL  frontal  side,  yellow 
arrow on the picture below.  
 

 

Milestone suggests freezing samples at -40°C. 
The freezing time depends on mould size used. 
Refer to chapter 3.10.1.
