Troubleshooting
This chapter provides solutions to performance and technical problems that may arise.
For each identified problem alternative actions are written as bullet points, and the first
action should be tried before moving onto the next .

Repack the column with a lower Packing
Factor.
Bed packed too densely.
Leading peak
•
Compress the bed further and retest
before repacking the column with a
higher Packing Factor.
Bed not packed densely
enough.
Tailing peak
•
Repack the column with a lower Packing
Factor.
Cracks in the bed due to a
too densely packed bed. The
cracks are mainly vertical.
Double peaks. The
'extra peak' is before
the main peak (i.e.
leading)
•
Try to compress the bed slightly and
retest. Otherwise, repack the column
with a higher Packing Factor.
Cracks in the bed or a gap
between the chromatogra-
phy medium and adapter.
The column may have been
packed too loosely.
Double peaks. The
'extra peak' is after the
main peak (i.e. tailing)
•
To remove protein:
Clean-in-place according to instructions.
You may need to extend the time re-
quired.
•
To remove chromatography medium:
For stainless steel bed support: Disas-
semble the column and remove the
medium using ultrasound or com-
pressed air, or replace the bed support.
For plastic bed support: Discard the
clogged PE bed supports.
Blocked bed support creates
zones of different liquid
velocities.
