GRADIENT CALCULATOR
The gradient is set within the program (see page 35 for further details), however, it is not possible to
view the actual temperatures of each column in the program. The gradient calculator calculates the
temperatures for each of the 12 columns (24 for the 384 well block) and allows the user to determine
the optimal amplification conditions for the assay.
The Gradient Calculator is found in the
System User menu.
Selecting "Temperature" enables the
temperature of the middle of the gradient to
be set. The gradient can be set between 20
and 70°C.
Selecting "Gradient" allows the range of the
gradient to be set. The minimum is 1°C and
the maximum is 30°C.
An example for the 60 x 0.5ml block is
shown. The gradient is 10°C with the middle
of the block set to 55°C, so the block will be
approximately 50°C on the left and 60°C on
the right and 55°C in the middle of the block.
It is possible to print the column and gradient
information by selecting "Print". Selecting
"Exit" returns the screen to the System User
menu.
