Chapter 5: Bidirectional Amplicon Tagging on the LP 48.48 IFC
Load the IFC

Vortex the sample mix solution for a minimum of 20 seconds and centrifuge for
30 seconds.
IMPORTANT
It is essential to vortex all components to ensure complete mixing.
Load the IFC

Pipet 4
μ
L of 20X primer solution into each of the primer inlets.

Pipet 4
μ
L of sample mix solution into each of the sample inlets.
IMPORTANT
While pipetting, do not go past the first stop on the pipette. Doing so may
introduce air bubbles into the inlets.
NOTE
An 8-channel pipette is recommended to load the sample mix and 20X primer
solutions. The recommended pipetting order is shown below.

Load the IFC into the
pre-PCR
IFC Controller AX in the pre-PCR lab.

Press
Eject
to move the tray out of the IFC Controller AX.

Place the IFC onto the tray by aligning the notched corner of the IFC to the A1 mark.

Press
Load Chip
to register the barcode of the IFC and activate the script selection.

Load the LP 48.48 IFC for Access Array. Select
Load Mix (155x)
and
Run Script.
NOTE
For Access Array 48.48 IFCs, select
Load Mix v7 (151x)
and
Run Script.
Load Mix
v7 (151x) is a script update from Load Mix (151x) for Access Array 48.48 IFCs and can be
downloaded from
fluidigm.com/software

for assistance.

Once the script is complete, press
Eject
to remove the IFC.
