b.
Remove all plastic and paper protections from each part of the microscope (eyepieces,
light, diaphragm,…)
c.
Familiarize with the mechanical components of the microscope. Move these
components to see how they behave and if they operate properly.
d.
Insert the plug into the socket at the back of the microscope. Connect the other end to
the electrical power supply. If the batteries are charged the microscope can be used
without connecting to the electrical power supply.
Note:
in case of using the microscope connected to the power supply, it should be properly
earthed. Make sure the input voltage is the appropriate for this equipment (100-240V; 50-
60Hz).
4.2 Using the instrument
a.
Turn the power source of the microscope on by pressing the switch at the back of the
microscope. The light intensity can be regulated by means of the knob placed at the
left side of the base of the microscope. It is recommended to be used at 70% of
intensity.
b.
Place the simple (microscope slide) and hold it with the clamp; to open the clamp use
the metal lever at the left side.
c.
The amplitude of the light beam that reaches the simple can be adjusted by changing
the diaphragm aperture. The maximum resolution is obtained by adjusting properly
this aperture. Every time the objective is changed and to get the best resolution it is
advisable to remove the eyepiece to see the size of the diaphragm aperture through
the eyepiece tube. The diaphragm aperture must be adjusted until it is a little smaller
than the objective aperture.
Note:
The diaphragm aperture does not change light intensity; for this use the knob
placed at the left side of the base of microscope.
d.
If needed put the filter in the filter holder.
e.
Start observation always with low magnification objectives 4x and 10x. To avoid the
objective touching the sample, turn the coarse focusing knob to lift the mechanical
stage until it reaches a few millimeters from the objective. The turn slowly the fine
focusing knob to focus the image.
f.
Once the image is well focused change to the next higher magnification objective and
adjust focus by turning only fine focusing knob. To change the objective turn the
nosepiece until hearing a “click” sound and make sure it is well centered in the light
beam. Always try to change the objective by holding the nosepiece not the objective
