method development is largely dependent on testing flow rates for different protocol solvents,
using the SPE consumables desired, and observing the flows visually.
You can execute the SPE method building process from the Otto SPEcialist's Manual Profile
control screen.
5.2 Developing an SPE method
Most SPE methods contain a common sequence of steps, similar to the following:
1. Add a liquid to the SPE consumable.
2. Slide the SPE consumable underneath the pressure manifold.
3. Close the manifold onto the consumable stack.
4. Execute an appropriate pressure profile.
5. Open the manifold.
6. Slide the SPE consumable out from underneath the pressure manifold.
7. Add the next liquid.
8. Repeat for each method step.
5.2.1 Creating a pressure profile
The key to creating a quality method is creating an appropriate pressure profile for each step in
the method protocol. The Otto SPEcialist's Manual Profile function is the best way to accomplish
creating these pressure profiles. Perform the sequence below for each protocol step.
To create a pressure profile:
1. On the main Otto touchscreen, tap
Pressure Profiles
>
Manual Profile
.
2. Add the appropriate liquid for the step in the method protocol to the SPE consumable.
3. Slide the consumable underneath the pressure manifold.
4. Establish a time scale of approximately two minutes by tapping the
Right Arrow
and
Left
Arrow
, as necessary. In most cases, the pressure profile for a given method step is a
much shorter amount of time. However, to allow sufficient time for the experiment, it is
better to err on the longer side during development. You can always shorten the profile
later.
5. Tap
Close Manifold
. The manifold lowers and seals on top of the consumable stack.
6. To activate the time, tap
Start
.
Notice:
When establishing a desired flow rate through the SPE column/plate,
do not increase the pressure too quickly. If the flow through the consumable is
too fast, you may need to repeat the experiment.
