Select from the list the range of wavelengths (
Emission
Bandwidth
parameter) centered around the emission wavelength
that will be detected. Two choices are available: 15 or 30 nm.
Enter the
Signal Offset
value (from 0 to 1000 mV) that will be
added to the detector output signal to create a positive baseline
offset.
Check
Shutdown lamp at sequence end
to switch off the lamp, at
the end of acquisition (Quick Start and sequence).
Check
Lock detector keyboard during runs
to lock the detector
keyboard.
Program the evolution of the
Excitation and Emission
wavelengths. The
Excitation
parameter is the wavelength of light
(from 200 to 850 nm) used to energize the molecules in the
column eluent. The
Emission
parameter is the target
fluorescence wavelength (from 250 to 900 nm) measured by the
PMT to generate the detector signal.
Check the
Az
parameter to zero the detector output at the
programmed step time.
