Problem

Hard to remove combs
Agarose too cold
Combs not removed immediately
after a gel has solidified
Remove Combs after a gel has
been allowed to solidify for 15
min. Removal at a later time
may cause the gel to adhere to
the combs or trapping of air
bubbles beneath the gel.
Difficult to visualize bands
Incorrect background color
Use a gray or black background.
Excessive leaking of samples
when a multi-channel pipette is
used
Leaking tips or tips not aligned
Make sure all pipette tips are
firmly attached by turning and
pushing tips against the nozzle,
one by one. Align tips before
use. Use non-autoclaved tips,
because autoclaving tends to
distort the shape of the tips.
