2) OBSERVATION IN PHASE CONTRAST

1.
Center the condenser as described on
page 25.
2.
Move the condenser slide to insert the
“10” position
3.
Insert the 10x objective into the optical
path.
4.
Open the aperture diaphragm.
5.
Place a sample on the table and focus.
6.
Remove an eyepiece and insert the
centering telescope (Fig.32)
7.
Rotate the top of the telescope to focus on
the rings (one light and one dark) visible in
the telescope. (Fig. 33)
8.
Using the centering screws on the slide

center the rings so that the clear ring

is
concentric to the dark ring
③
.
9. Repeat the operation with the 40x
objective: 40x objective - slide position
“40”.
10.
At the end remove the centering telescope,
reposition the eyepiece and start the
observation.
•
With the 40x objective, it may be useful
to slightly raise the condenser, to
obtain a better projection of the phase
rings. This is not a defect.
