Visually inspect the flow cell for bubbles passing through the lanes and leaks near
the manifolds.

Remove the waste tubing for the appropriate flow cell from the waste container. Make
sure that you do not include the waste tubing for the opposite flow cell, or the tubing
from the condensation pump or the priming pumps.

Bundle the waste tubing with parafilm, making sure to keep all of the ends even. Place
the bundled tube ends into a 250 ml bottle.

When the wash is complete, measure the delivered volume.
NOTE
The priming pump is washed during the wash steps. Keep the waste from the priming pump
separate.
The following tables list the wash volumes collected from the waste tubing. They do
not include the priming pump wash volume.

Load the instrument with 1 N NaOH as follows:
a
Load 8 SBS bottles with 5 ml of NaOH.
b
Load 10 PE tubes with 5 ml NaOH.

Load the bottles and tubes onto the instrument in the appropriate reagent rack.
