Preparation

Solution

Mix 150 ml ethanol with distilled water.

Make up to 1.0 l with distilled water.

Note:

Washing solution can be stored at room temperature for up to six
months.

Washing

Add 10 g citric acid to 500 ml distilled water and adjust to pH 2.3.

Make up to 1.0 l with distilled water.

Note:

Gels should be stored in storing solution at 4°C in a dark location or
away from light.

Deep Purple staining protocols

Action

Step

Fixing: Fix gels from 90 minutes to overnight in fixing solution.

Staining: Stain for 90 minutes at room temperature covered from
light using the prepared working staining solution. Prior to diluting
with borate buffer, ensure that the Deep Purple concentrate is
at room temperature.

Washing: Wash for 90 minutes in washing solution.

Acidifying: Acidify gels for 45 minutes. Further washes in this
solution can be performed if desired.

Imaging: Imaging is best performed using laser scanning
fluorescence flat bed imaging systems, such as the GE Typhoon™
FLA laser scanners.
