Vol. (Volume):
The total volume after filling samples and reagents in
the corresponding wells. The volume should be less than 1000μL to
avoid spilling.

Mix Time:
The time for mixing during this step
•
Mix Speed:
Higher the speed level, the faster the stirring. Generally,
high-viscosity liquids require high speed while low-viscosity liquids require
low speed, otherwise the liquid may splash.
•
Magnet Lowering (Time):
It is the time set for the magnetic rod head to
move from the liquid level towards the bottom of the well at a constant
speed. Generally, it should be more than 10 seconds, which is good
for collecting magnetic beads. But don
’
t set it too long, otherwise the
beads may stick to the side of the well. Different experiments may
require different lowering time.
•
Magnet Resting (Time):
It is the residence time of the magnetic rod head
after moving to the bottom of the wells. Generally, this
time should not
be less than 1 second, and it is determined by experiment.
•
Collecting Cycles:
The number of times of "Magnet Lowering" and
"Magnet Resting". Multiple times of magnetic bead collection can
bind more magnetic beads.
• If the step does not require magnetic bead collection, the number of
"Collecting Cycles" can be set to "0". For example, after elution the
beads are deposited back to the initial well (well 2/8) and doesn
’
t need
to be collected. In this step the "Collecting Cycles" can be set to "0"
to separate the magnetic beads from the sleeves (only the magnetic
rod moves up).
