1)
Attach the coupling to your microscope
Cairn microscope adapters are either supplied to couple onto an existing epi-illuminator or are used in
place of the epi-illuminator as a ‘direct’ coupling.
a.
If attaching to your existing epi-illuminator, a male bayonet coupling is normally supplied,
which attaches to your microscope epi-illuminator via a large grub screw.

Align and focus the 340nm LED at the back focal plane of the objective
** IMPORTANT SAFETY NOTE - UV LEDs ARE NOT VISIBLE BY EYE AND WILL CAUSE DAMAGE IF
LOOKED AT DIRECTLY.
ALWAYS VIEW THE LED LIGHT BY SHINING AT A PIECE OF WHITE PAPER AND VIEW THE
FLUORESCENCE AT A LOW POWER **

Move to a blank objective position on the objective nosepiece, remove any cover caps and
place a small piece of white paper in this position. Alternatively, place a centering tool in the
light
path.

Ensure your Fura-2 cube is in place in your microscope.
c.
Turn on the 340nm LED channel on the OptoLED controller
d.
Using the 2.5mm allan key (provided), turn the
340nm LED focus
until the image of the
LED comes into sharp focus on the paper, please refer to Figure 1 below.

Using the same 2.5mm allan key, centre the position of the 340nm LED using the
340nm

Repeat the steps above for X, Y and Z adjustment for the 385nm LED
Once the FuraLED channels have been aligned and focussed, the coupling should not need further ad-
justment unless removed from the microscope. We recommend periodically checking the alignment to
ensure your illumination is optimal.
