MICROSCOPE
Prepare a “slide” for wet mount observation
Learn this technique so that you can then apply it to the many observations of “slides” (plastic) with living organisms, or
those that cannot be stored (e.g.: cells, algae, starch granules, etc.).
Material
• a sheet of white paper
• a black cardboard
• plastic “slide”
• “slide” coverslip (you can cut it from the plastic sheet in your kit)
• test tube with pure water
• pipette
• filter or absorbent paper
• plastic stick
• tray (to contain and wash the used “slides” with a drop of dishwashing liquid)
Operation

You will work best on a rigid surface that is firm and sturdy, covered with a sheet of white paper
and a black card in a corner, in order to have a dark background for transparent objects that are
free of contrast.

Use the plastic stick or the pipette to withdraw a small drop of liquid
containing the object you’ll examine and place it on the slide, or,
after putting a drop of water on the slide, place the material you will
examine in it.

Apply a coverslip as shown.
NOTE:
in the next few observations with your microscope, this symbol will be used to indicate how to
prepare the slide.

If you need to add a liquid or a dye that you
have prepared, put a drop on one side (on the left, for example)
of the cover slip and put a piece of filter or absorbent paper on the
opposite side (right), which will attract and absorb the drop.
ONION SKIN CELLS
This is one of the most fun observations and easy to do.
It is an observation with slides.
1)
Find an onion and detach a thin transparent layer from the convex part
2)
Use tweezers to transfer a portion of the epidermis on the slide and
make an observation.

OTE:
for this observation, you may need to also light the
slide from underneath in order to create a greater contrast
and to view all the cells on the screen. You can simply use a
penlight and point it toward the slide.
